Argireline
In a clinical study published in the International Journal of Cosmetic Science, Argireline was shown to reduce wrinkles up to 30% after only 30 days of use. Argireline reduces the depth of expression wrinkles, especially in the forehead and around the eyes. Argireline is a safer, cheaper, and milder alternative to Botox®, topically targeting the same wrinkle-formation mechanism in a very different way.
Angireline works by reducing the depth of wrinkles on the face caused by the contraction of muscles that cause facial expression, especially where they are most prominent, such as the forehead and around the eyes. Studies also showed Angireline can prevent apparent aging in the skin cause by repeated facial expressions, and the release of a substance called catecholamine inside the body that leads to skin aging.

Argireline Efficacy (Effectiveness) Testing
The anti-wrinkle effect of Argireline was ascertained in two different in vitro tests directly related to the formation of wrinkles in the epidermis as well as a separate in vivo test performed on healthy human volunteers.
About Argireline
Argireline’s specific sequence was discovered within a combinatorial library of hexapeptides developed by Lipotec, SA (Barcelona, Spain). Several proprietary processes led to the identification of this particular acetylated hexapeptide, which was shown to inhibit the SNARE complex formation and to also inhibit catecholamine release in high throughput screening conducted on chromaffin cells.
Once identified, the biologically active hexapeptide was synthesized by solid phase peptide synthesis using the Fmoc/tBu protocol. The peptide was purified and characterized by amino acid analysis, mass spectroscopy and NMR. Homogeneity of the peptide was assessedby HPLC and capillary electrophoresis.
Argireline was found to inhibit vesicle docking by preventing the formation of the essential ternary SNARE Complex. Inhibition of noradrenaline and adrenaline release was also demonstrated in a second in vitro study supporting the anti-wrinkle activity observed in a thirty (30) day clinical study. In vivo tests further demonstrated the benefits of Argireline. Skin topography analysis (for measuring the effectiveness of an O/W emulsion containing 5% Argireline) was performed using silicon replicas from around the eyes of ten healthy female volunteers. Replicas were obtained at 0, 15 and 30 days of a twice-a-day treatment regimen. Analyses of the imprints were performed by confocal laser scanning microscopy to assess the evolution of the skin surface before and after treatment. Skin topography images from the three dimensional reconstruction of optical sections are available upon request. It was observed that the severity of wrinkles around the eyes decreased up to 17% after 15 days of treatment and up to 27% after 30 days of treatment, substantiating the proposed biochemical mechanism hypothesis.

During the aging process, the synthesis of collagen is reduced and both degradation and disorganization of the fibril network are increased. Trylagen acts at these three stages in the life of collagen and its activity is made up of three main functions: (1) boosts the synthesis of Collagen types I, III and IV, (2) controls collagen fibril dimensions and inhibits enzymatic destruction, (3) avoids excessive collagen damage in aged skin.
Clinical tests have shown the following effects of using Tryalen:
- 128% increase of Collagen I synthesis after 15 days: Type I collagen fibrils have a great tensile strength and elastic resistance
- 81% increase of Collagen IV synthesis after 15 days: Type IV collagen is the most abundant structural component of basement membranes
- Collagen III production tripled at the highest dose: Youthful skin contains a predominance of collagen III, but during the aging process, cells gradually lose their ability to produce this type of collagen

6.0 GATULINE®

DETAILS
GATULINE is extracted from the upper parts of the Acmella oleracea plant belonging to the Compositæ family (Compositæ-Heliantheæ). This small plant, which blossoms all year long, has a broad geographical footprint, covering the whole tropical zone in South America, Africa and Asia.
In-Vivo Studies (on human volunteers)
The in vivo smoothing efficacy of GATULINE has been demonstrated on “crow’s feet” wrinkles. Skin smoothing was assessed by topometric analysis using a hi-tech technique: the Fringe Projection technique. This allowed GATTEFOSSÉ, who conducted the test, to demonstrate very precisely the benefits provided by daily application of GATULINE.
Fringe Projection, a purely optical method used to assess roughness of the skin, was conducted on two panels of volunteers (20 and 30 people) with twice-daily application for 28 days:
- 1 panel using a placebo formulation on half of the face and a formulation containing 2% of GATULINE on the other half
- 1 panel using a placebo formulation on half of the face and a formulation containing 5% of GATULINE on the other
Over 75% of these test subjects reported that their wrinkles decreased one day after use on the side of their face that used GATULINE.
In-Vitro Studies (in Test Tubes)
The myorelaxing activity (muscle relaxing) of the extract was first tested on a model reproducing muscle contractions. This model is produced by the co-culture of motor neurones with human muscle cells forming striated muscle fibres which, once innervated, spontaneously contract. The muscle contractions were counted to prove that GATULINE extract blocked these contractions. This blocking process is fully dependent on the concentration of the tested extract. As such, it was demonstrated that just 0.6% of GATULINE is enough for complete myorelaxation.
GATULINE shows a fully reversible function (complete recovery after 24 hours) and is thus positioned as a very safe extract while remaining highly effective.
The efficacy of standard formulations (O/W emulsions) containing GATULINE was evaluated against a placebo. Muscle contractions were counted to highlight the activity of creams containing GATULINE on human skin and cells.
